Amphotericin B is a polyene antifungal. It binds ergosterol in the fungal cell membrane and forms pores that make the cell leak and die. It is one of the oldest and broadest-spectrum antifungals in use.
It is approved for serious, potentially life-threatening fungal infections, including cryptococcal meningitis, invasive candidiasis, aspergillosis and mucormycosis, and the liposomal form is also approved for empirical treatment of presumed fungal infection in febrile neutropenic patients and for visceral leishmaniasis. Amphotericin B comes in different formulations: conventional amphotericin B deoxycholate and lipid formulations such as liposomal amphotericin B (originator brand AmBisome). The formulations are dosed differently and are not interchangeable. It is on the WHO Model List of Essential Medicines.
