Dacarbazine, also called DTIC, is an alkylating agent. It is activated in the liver to a compound that adds methyl groups to DNA, damaging the DNA of cancer cells; temozolomide works through the same active compound.
It is approved for metastatic malignant melanoma and, in combination with other medicines, for Hodgkin lymphoma, where it is part of the ABVD regimen with doxorubicin, bleomycin and vinblastine. In some countries it is also approved for advanced soft tissue sarcomas. Dacarbazine is on the WHO Model List of Essential Medicines.
Dacarbazine is given by intravenous infusion in hospital and handled under cytotoxic safe-handling procedures.
