Midostaurin is an oral multikinase inhibitor. It blocks FLT3, which is mutated in about a third of acute myeloid leukaemias, and KIT, including the D816V mutant that drives systemic mastocytosis.
It is approved for newly diagnosed acute myeloid leukaemia with a FLT3 mutation, in combination with standard induction and consolidation chemotherapy, followed in Europe by single-agent maintenance; and for advanced systemic mastocytosis, including aggressive systemic mastocytosis, systemic mastocytosis with an associated haematological neoplasm and mast cell leukaemia. FLT3 status is confirmed with a validated test. The originator brand is Rydapt.
Midostaurin soft capsules come in blister packs; nausea and vomiting are common, and haematologists use preventive anti-sickness medicines.
