Pomalidomide is an immunomodulatory medicine, a structural analogue of thalidomide and lenalidomide. It acts on the immune system and on the bone-marrow environment that supports myeloma cells.
It is approved for relapsed or refractory multiple myeloma in patients who have received prior treatment including lenalidomide, in combination with dexamethasone, and in Europe also with bortezomib and dexamethasone. In the United States it is also approved for AIDS-related Kaposi sarcoma, and for Kaposi sarcoma in HIV-negative patients. Because of the risk of birth defects it is supplied under a pregnancy prevention programme in many countries. The originator brand is Pomalyst (Imnovid in Europe).
