Ribociclib is a cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor. Blocking these kinases stops hormone-receptor-positive breast cancer cells from moving through the cell cycle and dividing.
It is approved for hormone-receptor-positive, HER2-negative advanced or metastatic breast cancer, in combination with an aromatase inhibitor or with fulvestrant, in postmenopausal women and, with a GnRH agonist, in pre- and perimenopausal women and men. It is also approved, with an aromatase inhibitor, as adjuvant treatment of hormone-receptor-positive, HER2-negative early breast cancer at high risk of recurrence. The originator brand is Kisqali.
Ribociclib can prolong the QT interval, so ECGs and blood tests are done during treatment.
