About tegafur, gimeracil and oteracil: drug class and approved uses
Tegafur, gimeracil and oteracil, often called S-1, is an oral fluoropyrimidine combination. Tegafur is a prodrug of fluorouracil; gimeracil slows the breakdown of fluorouracil so that levels stay higher; and oteracil reduces the activation of fluorouracil in the gut lining, to lessen gastrointestinal side effects.
In the European Union the combination is approved, with cisplatin, for advanced gastric cancer in adults, and for metastatic colorectal cancer in patients who cannot continue other fluoropyrimidine treatment because of hand-foot syndrome or cardiovascular toxicity. In Japan and several other Asian countries it has wider approvals, including gastric and other gastrointestinal cancers. The originator brands are TS-One in Japan and Teysuno in Europe. It is not approved in the United States.
