Memantine is a low-to-moderate affinity, uncompetitive NMDA receptor antagonist. In Alzheimer's disease it is thought to protect nerve cells from the effects of persistently raised glutamate while still allowing normal signalling.
It is approved for moderate to severe dementia of the Alzheimer's type. It can be used on its own or together with a cholinesterase inhibitor such as donepezil, and a fixed-dose combination with donepezil is approved in the United States. Memantine treats symptoms; it does not change the underlying course of the disease. The originator brands are Ebixa in Europe and Namenda in the United States.
