Nintedanib is an intracellular tyrosine kinase inhibitor that blocks the VEGF, FGF and PDGF receptors. In the lungs this slows the activity of fibroblasts, the cells that lay down scar tissue.
It is approved for idiopathic pulmonary fibrosis (IPF), for other chronic fibrosing interstitial lung diseases with a progressive phenotype, and for interstitial lung disease associated with systemic sclerosis. Approvals for children with fibrosing ILD have also been granted. The originator brand for these uses is Ofev. Separately, under the brand Vargatef, nintedanib is approved in Europe with docetaxel for a type of advanced non-small cell lung cancer.
Liver function is monitored during treatment, and diarrhoea is a common side effect that prescribers manage.
