Prucalopride is a selective, high-affinity serotonin 5-HT4 receptor agonist. Stimulating 5-HT4 receptors in the gut wall increases the coordinated contractions that move contents through the colon. Its high selectivity distinguishes it from older 5-HT4 agonists that were withdrawn because of effects on the heart.
It is approved for chronic idiopathic constipation in adults; in Europe, specifically for adults in whom laxatives fail to provide adequate relief. It is not approved for constipation caused by medicines or by other diseases in most labels. The originator brands are Resolor in Europe and Motegrity in the United States.
